T47D Cell

T47D cell is continuous cell line isolated from ductal tumor of a woman breasttissue aged 54 years old. Continous cell line is often used in cancer research in vitro because several reason, such as easy to handle, has infinity replication function, high homogeneity, and easily replaced with a frozen stock in the event of contamination (Burdall et al., 2003). The morphology of T47D cell is like the epithelial cell. This cell cultures in DMEM + 10% FBS + 2 mM L-Glutamine and incubated in CO2 5% incubator and 37oC in themperature (Abcam, 2007).

T47D cells expressed mutated p%3 protein. Missence mutation happens on 194 residues ( on Zinc-binding domain, L2), so that p53 can’t bind the response element in DNAThis results in reduced even loss of the ability of p53 to regulation of cell cycle. T47D cells is ER/PR positive breast cancer

Selkankerpayudara T47D mengekspresikan protein p53 yang termutasi.Misssence mutation terjadipadaresidu 194 (dalam zinc-binding domain, L2), sehingga p53 tidakdapatberikatandengan response element pada DNA (Schafer et al., 2000). The induction of exogenous estrogen results on the increase of proliferation proccess (Verma et al., 1998). The T47D cell is the cell sensitive with doxorubicin exposure (Zampieri et al., 2002).
